A histological examination of grafting success in pearl oyster Pinctada margaritifera in French Polynesia ArchiMer

Pearl oyster grafting is a complex surgical operation that should lead to pearl formation after approximately eighteen months. Although this technique has been used for many years in French Polynesia, the grafting process is still not standardised. While studies have been carried out in order to improve graft performance and yield, these remain highly variable due to post-grafting mortality, nucleus rejection and unreliable pearl quality, all of which constrain pearl farm profitability. The present study uses histological analysis to monitor oysters that either rejected or retained their nuclei. Bilan 2005-2006 du Réseau de Veille Zoosanitaire des huîtres perlières, Pinctada margaritifera, en Polynésie française ArchiMer

En 2005 et 2006, un total de 1333 analyses réparties en 1109 huîtres perlières, P. margaritifera, 93 pipi, P. maculata, 27 chama, Chamidae sp, 46 kapi kapi, Saccostrea cucullata et 58 Isognomon sp. ont été effectuées dans le cadre du REseau de PAthologie des huîtres perlières (REPANUI). Les analyses histologiques ont révélé comme les années précédentes la présence de sporozoaires de type grégarine, des parasites de type Rickettsien, des kystes de cestodes (Tylocephalum sp.). Biofouling development and its effect on growth and reproduction of the farmed pearl oyster Pinctada margaritifera ArchiMer

In bivalve aquaculture, dominant fouling organisms are filter feeders which can compete for food with reared bivalves, sometimes causing mortality or reducing their growth rate. This study investigated the effect of biofouling on the farmed pearl oyster Pinctada margaritifera in two lagoons of French Polynesia. Survival, growth and reproduction of 2 year-old pearl oysters were monitored with regular sampling schedules, from the initial stage of colonization up to 20 months of biofouling accumulation. A transplant is a complex surgical procedure performed to obtain a pearl after about 12 to 18 months. One of the key steps in this procedure is the transfer of the graft (piece of mantle) from the donor oyster. A functional relationship exists between the coating nature of the pearl and the cellular structure of the pearl bag portion of the graft. We have identified two genetic markers of biomineralisation in Pinctada margaritifera, the pearl oyster: Perline and Calcine, responsible for secretion of aragonite and calcite, respectively. Development of an antiseptic procedure to improve retention rates during the grafting process of black-lip oyster, Pinctada margaritifera ArchiMer

In French Polynesia, black pearl cultivation now takes place on about 30 of the 118 atolls or islands. This activity has largely replaced copra, coconut oil and fishing as a source of income. Around 1,000 pearl farms, generally small ones of less than 5 ha, are registered. The pearl farms are mainly owned by families. In 1998, about 7,000 persons were employed in theentire pearl industry, from production to retailing. In value, this activity represents the first exportation of French Polynesian products. From 1988 to date, exports increased exponentially before leveling. Starting with less than two kilograms in 1972, exports have now stabilized at Il tons of "black" pearls.

Effect of food conditioning on gonadic activity in the oyster Pinctada margaritifera ArchiMer

Preliminary experiments allowed defining basic requirements of Pinctada margaritifera for somatic and gonadic growth in controlled conditions. This study was continued by observation of the gametogenesis at a cellular level. In April/May 2008, 125 oysters were conditioned for 30 days in 25 tubular tanks (30 L). Shell growth, reproduction, and natural mortality of the giant clam Tridacna maxima were characterized over a two-year-period in the lagoon of the high island of Tubuai (Austral Archipelago) and in the semi-closed lagoon of Tatakoto (Tuamotu Archipelago) in French Polynesia. We also recorded temperature, water level, tidal slope, tidal range, and mean wave height in both lagoons. Lower lagoon aperture and exposure to oceanic swells at Tatakoto than at Tubuai was responsible for lower lagoon water renewal, as well as higher variability in temperature and water level at Tatakoto across the studied period. Proteinaceous exotoxins of shrimp-pathogenic isolates of Vibrio penaeicida and Vibrio nigripulchritudo ArchiMer

The pathogenicity of two V. penaeicida strains, AM101 and KH-1, with different geographic origin, and V. nigripulchritudo strain AM102, were investigated in juvenile blue shrimp species Litopenaeus stylirostris. Alive bacteria and protein fractions (PFs) obtained from cell-free supernatants (CFS) were used in experimental challenges. Strains AM102, AM101, and KH-1 produced respectively 60, 54 and 12% mortality at 96h after infection using 104 cfu mL-1 of bacterial suspension. Exocellular toxin-like factors were evidenced in CFS from the New Caledonian strains (AM102 and AM101) but not the Japanese strain (KH-1). Rapid and sensitive PCR detection of Vibrio penaeicida, the putative etiological agent of syndrome 93 in New Caledonia ArchiMer

Experimental infections of Penaeus (Litopenaeus) stylirostris were performed with a Vibrio penaeicida strain (AM101) isolated in New Caledonia from Syndrome 93 diseased shrimp. Cumulative mortalities resulting from intramuscular injection or immersion of shrimp in bacterial suspensions demonstrated high virulence for this bacterial strain and suggested that V. penaeicida could be the etiological agent of Syndrome 93. La mise en place d’un réseau d’observation de la qualité des eaux lagonaires de Polynésie française, financé par le contrat de projet Etat – Pays, a été initiée mi 2012 pour une durée de deux ans. Durant un à trois semestres la contamination in situ a été suivie sur 6 sites : Vairao et la baie de Phaéton à Tahiti, Moorea (Vaiare), Huahine (Faie), Maupiti et Rangiroa. Le mollusque bioindicateur utilisé est l’huître perlière Pinctada margaritifera. Après transplantation et 4 mois d’immersion sur site, la mesure dans les chairs de la radioactivité gamma n’a pas permis de détecter de radionucléides artificiels, cependant du césium 137 a été mesuré dans les sédiments sur 2 sites.
